Loki, Yep you got it right. All that's required to keep the Mom's happy is to re-tube with fresh medium every 30 days or so.
If your not gonna use a mom for a specific cycle just cut off a fresh shoot from mom and re-tube,trash what's not needed, she's good to go and be maintained for another cycle. No worry about generation decline, since it's the same genetics.

How fresh of a sample do you need, to culture some cells?

Can it be from a day old cutting?

Well, the fresher the better. I'd think a day or two would be ok, depending how the cutting was handled. If you put it in some water, like a vase, and kept in a cool area, you'd be able to get viable cells for at least a week or more. Technically, you only need 1 cell. I've gotten good onion cells after the onion was cut in half and stored in the fridge for at least a week or more.

Thanks for the offer Earl, but I seem to have enough for now. She looks pretty tempting in your photo. Speaking of photo, here is a shot of the PPP at 8 days into this. I'm not seeing any contamination as of yet and I think she's starting to grow some.

This is a shot of the AK at 8 days. Of all of the cultures I took during this first attempt. 16 of the pot starts, 8 African Violet, 1 Fern, and dividing some Iris, 12 of those, I don't see anything bad growing in any of the tubes or jars. Just lucky I guess. I would of thought that by now any contamination would have shown up. Just waiting and watching things grow.

Thanks Earl, I'll give them a try. I've been wanting to get some Blueberry going again. I love how long the buzz lasts and it takes the pain down a notch for a good long time. I let my Mother go a couple of years ago and haven't had any since.
I'm wanting to get that and Northern Lights in culture, along with the White Widow, AK, and PPP that I already have. I let my N. L. Mom go a couple of years ago also and I want her back now I can keep more strains going.
I've been out of town for a couple of days and haven't taken a look at the micro grow for a few days so there might be some pics. in the next post.
